Nata a Scottsdale in Arizona da Jeff e Krista Stone, ha un fratello minore di nome Spencer. Membro del Valley Youth Theatre, teatro regionale di Phoenix, appare all'età di 11 anni nel musical The Wind in the Willows e successivamente in A Winnie-the-Pooh Christmas Tail, La principessa sul pisello, Cenerentola, The Wiz, Titanic, Honk!, La sirenetta, Schoolhouse Rock Live!, Alice nel Paese delle Meraviglie e in Joseph and the Amazing Technicolor Dreamcoat. All'eta di 15 anni si trasferisce con la madre a Los Angeles in California. Nel 2004 comincia la sua carriera televisiva, appare in numerose serie telivisive, The New Partridge Family, Medium, Zack e Cody al Grand Hotel, Malcolm, Lucky Louie, Drive. Ha fatto il suo film d'esordio nel 2007 con la commedia Suxbad - Tre menti sopra il pelo. Raggiunge la notorietà nel 2008 con i film The Rocker - Il batterista nudo e La coniglietta di casa con Anna Faris. Nel 2010, la Stone presta la voce al personaggio Mazie in Sansone, ed ottiene la parte della protagonista nella commedia Easy Girl, che le vale una nomination per al Golden Globe come migliore attrice in una commedia e una vincita agli MTV Movie Awards 2011 nella categoria miglior performance comica. Nel 2012 la vedremo vestire i panni di Gwen Stacy nel reboot di Spider-Man dal titolo The Amazing Spider-Man.
